8） 放置于30℃培养箱或者水浴中孵育30 分钟,15 分钟时混匀一次。
9） 30 分钟后，每管加入15μl DMSO(DIMETHYL SULFOXIDE)，轻轻上下颠
Bait 基因的自激活检测
一般采用膜检的方法，检测LacZ 基因的表达情况。膜检方法如下:
1) 将阳性克隆菌落影印于滤纸上。
2) 将滤纸完全浸于液氮中，时间长于30 秒，取出后晾干。
3) 培养皿加入适量显色液(新鲜配制)，垫一层滤纸，让其完全湿润，将冻溶
后的滤纸铺在上面，使显色液渗透到表层。9cm 培养皿加2ml 显色液，15cm
培养皿加5ml 显色液。
4) 盖上皿盖，放置在37℃培养箱中，避光放置。
5) 20 分钟后开始观察有无变蓝的阳性结果并记录，一般以3 小时为准，特殊情
况下可以看过夜是否变蓝。
6)反应过后打开皿盖，将滤纸烘干，保存并记录。
